Human intercostal skeletal muscle aging atlas (scRNA-seq and snRNA-seq, young vs aged)
A total of 43 coarse and 82 fine-grained skeletal muscle cell type clusters were identified from the joint single-cell (90,902 cells) and single-nuclei (92,259 cells) skeletal muscle atlas from 17 deceased transplant organ donors (8 young, ~20-40 y; 9 aged, ~60-75 y). Cells and nuclei were first integrated separately with scVI using 10X run as a batch and donors and 10x Genomics Chromium chemistries (v2, v3 or v2/v3) as categorical covariates. They underwent iterative leiden clustering and were initially annotated using cannonical markers in the field and later in-house generated celltypist model trained on the earlier version of the dataset.
